Not sure if this applies to Vista or just Windows7, if you use bitlocker to encrypt a usb drive each time you remove it you will have to type in your password, but there isnt a facility to re-lock internal hard drives even if you log out anyone else can access that drive unless you reboot or disconnect the drive, if you open a Run box and type in manage-bde -lock E: it will re-lock the drive, change the drive letter if yours is different.
